English: Division and migration of QL.a/p in wild-type. casIs330[Pegl-17::myr::mCherry, Pegl-17::HIS-24::mCherry, Pegl-17:: MOEabd::GFP] was used to image QL division and QL.a/p migrations. mCherry labels cell membranes and chromatin (red) and GFP labels F-actin (green). Images were acquired every 2 minutes using a spinning disk confocal microscope (Zeiss Axio Observer Z1, with Yokogawa CSU-X1 Spinning Disk Unit). Total imaging time was 250 minutes. Time-lapse imaging with casIs330 resulted in an approximately 2-fold delay in events relative to ayIs9 in timepoint analysis. Anterior is left, and dorsal is up. |
