Description
English: DNA replication always proceeds in the 5'-> 3' direction, by adding an incoming deoxyribonucleotide triphosphate (dNTP) to the 3' end of the existing strand. Here we see what would happen if we tried to add a dNTP to the 5' end of a strand. The reactive leaving group (2 extra phosphates) is on the 5' carbon of the dNTP, far from the 5' phosphate of the existing strand where the reaction would need to take place. Nucleophilic attack is not favored under these circumstances.
