Construite de 1867 à 1870, l'église Saint-Luke a été érigée pour la communauté anglicane de Waterloo. L'architecte Thomas Seaton Scott l'a conçue dans le style néogothique. Les proportions, la tour, les contreforts et les multiples ouvertures en ogive donnent au bâtiment un effet de puissance. Les vitraux commémoratifs qui ornent les fenêtres ont été offerts par des famille de Waterloo entre 1877 et 1962.

L'église Saint-Luke témoigne du courant néogothique qui a marqué au XIXe siècle l'architecture religieuse au Québec et elle serait la seule église québécoise conçue par l'architecte Scott.

L'église Saint-Luke a été classée monument historique le 26 juillet 1978 par le ministre des Affaires culturelles.
