Description
Deutsch: CRISPR-Cas9 kann als molekulares Werkzeug gezielte DNA-Doppelstrang Brüche einführen. Wenn die Sequenzen von tracrRNA und crRNA mit einem Tetraloop verbunden werden, entsteht eine single-guidance RNA (sgRNA), welche sich chemisch einfach synthetisieren lässt. So wird CRISPR/Cas9 zum einfach programmierebaren Werkzeug. Der Komplex aus sgRNA und Cas9 scannt intrazelluläre DNA nach einem Protospacer Adjacent Motif (PAM). Wird ein solches Motiv gefunden, wird die komplementäre DNA mit der crRNA-Region verglichen. Stimmen diese überein, so wird der DNA-Doppelstrang geschnitten. Die HNH Domäne schneidet den Ziel-Strang, während die RuvC-Domäne den gegenüberliegenden Strang schneidet.
