Deutsch: Zu A) Ohne angelegtes Feld sind die Ampholytmoleküle ungeordnet im Gel. Überall im Gel ist der pH-Wert identisch (z.B. pH = 7) Die Moleküle, die einen höheren isoelektrischen Punkt (pI) als der pH-Wert der Flüssigkeit besitzen, sind positiv geladen (q > 0). Diejenigen Ampholytmoleküle deren pI kleiner als der umgebende pH-Wert ist, besitzen eine negative Ladung (q < 0). Je größer der Unterschied zwischen pI und dem pH-Wert desto höher ist der Betrag der Ladung.
Zu B) Legt man eine Spannung an, so werden positiv geladene Moleküle zum (+)-Pol (Kathode) und negativ geladene Moleküle zum (-)-Pol gezogen. Es kommt zu einer Sortierung der Ampholytmoleküle nach ihrem pI. Am Ort der Aufkonzentration sorgen sie auch für einem pH-Wert der ihrem pI-Wer enmtspricht. Dadurch beeinflussen die Trägerampholyte auch die Molekülladung anderer Trägerampholytmoleküle. Insgesamt beginnt sich ein pH-Gradient auszubilden. Zu C) Die Moleküle konzentrieren sich dort auf, wo der pH-Wert ihrem pI entspricht. Dort sind sie dann nicht mehr geladen (q = 0). Insgesamt entsteht so ein sich selbst stabilisierender pH-Gradient. Diffundiert ein Molekül weg, so liegt es wieder geladen vor und wird durch den Strom zurück transportiert.English: A) Without electrical current the carrier ampholyte molecules are unsortet.
