Formación de gránulos de insulina en la red trans-Golgi.
La proinsulina se secuestra en el sitio de gemación del gránulo secretor a través de interacciones con proteínas como la cromogranina A/B (CgA/B) y VGF, y juntas forman grandes agregados.
Las proteínas unidas a la membrana como Phogrin y Secretogranin 3 (SgIII) proporcionan una clasificación adicional, y esto ocurre en paralelo con la clasificación de las proteínas procesadoras de proinsulina (PC1/3, PC2 y CPE).

La gemación de los gránulos de insulina depende de la acción de Arf1 y la gemación de la membrana se estabiliza con clatrina, el adaptador de clatrina AP-1 y las proteínas del dominio BAR (PICK1 e ICA69) y se ve facilitada por altas concentraciones del fosfolípido PI4P.
