English: "Adverse genetic effects of feeding high levels of a damaged nucleoside to mice. (a) Schematic illustration of oral feeding of dU to mice and analysis of intestinal tissue. LC–MS/MS quantification results of (b) dU and (c) 8-oxo-dG in gDNA extracted from the intestines of control (−) and dU-fed mice (+), showing 2.5-fold to 15-fold increases in levels of dU in the genomic DNA from these tissues. (d) Immunostaining of γ-H2AX in villi in the small intestine, showing enhanced DNA double-strand break (DSB) signals in response to dU feeding. Also shown are images of crypts in the large intestine. Tissues were costained with Hoechst 33343 (5 μg/mL) to highlight nuclear DNA. (e) Quantified intensities of γ-H2AX from red channels in panel d. Uncertainties are standard deviations (****p ≤ 0.0001) by the unpaired t test."
