Edificada nos séculos XV/XVI, destaca-se pelo seu belo pórtico manuelino, com colunas retorcidas que terminam em pináculos. Os portais laterais, mais simples, datam do mesmo período. Com interior de três naves. Os capitéis das colunas são formados por cordões torcidos, repetem o tema decorativo do portal principal. Na Capela-mor o retábulo de talha dourada (séc. XVIII) tem a originalidade de apresentar, no arco, dois anjos segurando a lua e o sol enquanto dois curiosos atlantes suportam todo o conjunto. Interessante sacrário em forma de templete. A imagem de Nossa Senhora da Conceição (sec. XVIII) é atribuída ao escultor Machado de Castro.

A capela do Santíssimo contém um pequeno tesouro de azulejaria do séc. XVII, constituído pelos que revestem a abóboda manuelina, que incluem 4 painéis de alminhas, e os das paredes historiano São Francisco salvando as almas e São Miguel matando o Diabo. O retábulo de madeira de castanho encerada (sec. XVIII) da capela de Nossa Senhora do Carmo, possivelmente proveniente do extinto convento franciscano, tem planta convexa, o que é pouco usual no Algarve. A igreja, para além de um bom núcleo de imagens, tem um tesouro sacro composto por objectos de culto que pertenceram ao antigo convento de Nossa Senhora do Desterro (sec.
