Live confocal microscope imaging of the lateral germinal crescent region of an HH4 [Tg(hUbC:H2B-Cerulean-2A-Dendra2)] quail embryo that was injected with an antibody against fibronectin (abB3/D6). The antibody was directly conjugated to AlexaFluor 594 (red). The 20x images are maximum intensity projections of 3 optical slices (2.3 μm each) taken from the dorsal aspect at 1.5 min intervals. Total elapsed time is 43 min. Cell nuclei = H2B-Cerulean (blue), cell bodies = Dendra2 (green). PGCs are larger and brighter than the surrounding tissue and appear in clusters and individually. Anterior to top, mediolateral midline to the right out of view. Embryonic tissue to the lower right. Extra-embryonic tissue to the left and upper left. Scale bar = 25 μm. |
