Description Video S8. Post-CSAT antibody injection movie for embryo 2. See Video S7 for pre-CSAT injection cell movements. CSAT antibodies against integrin β1 subunits fail to affect the cell motility and morphology of PGCs in HH5 embryos. Time-lapse confocal microscope imaging of the central germinal crescent region of an HH5 [Tg(hUbC:H2B-Cerulean-2A-Dendra2)] quail embryo that was injected with an antibody against fibronectin (abB3/D6). The antibody was directly conjugated to AlexaFluor 594 (red). After incubation, the germinal crescent region was injected again, this time with CSAT antibodies (174 ug/ml) and imaged live. The 20x images, with 2x optical zoom are maximum intensity projections of 3 optical slices (2.3 μm each) taken from the dorsal aspect at 1.5 min intervals. Total elapsed time is 24 min. Cell nuclei = H2B-Cerulean (blue), cell bodies = Dendra2 (green). PGCs are larger and brighter than the surrounding tissue and appear individually and in clumps in this movie. Anterior to the top. Embryonic tissue toward the bottom. Extra-embryonic tissue at the top. Scale bar = 25 μm.
