English: Analyses of mutant RAD18 localization during G1, S and G2 phase. Time lapse images of nuclei expressing YFP-RAD18 carrying a mutation in its Zinc finger (D221A). Images were captured every 30 min for 27 hours, a single nucleus was selected, aligned, and 5 frames of the images are shown per second. Endogenous RAD18 was downregulated by siRNA. Analyses started in G1 phase, and the cell went through S, and most likely G2. The cell cycle phases of mutant YFP-D221A were inferred from the time passed since the previous M-phase or the time span until the next M-phase, and the duration of G1, S, G2 and M phase in wild type cells. The growth rates of wild type and mutant cells were not different (data not shown). |
