Description
English: Supplemental video file for Figure 9 (M6PR and Stat2). Vero cells were infected with WT HPIV1 for 48 h, mock-stimulated with medium containing neither FCS nor IFN-β for 1 h and subsequently fixed, permeabilized and stained for nucleus (DAPI, blue), M6PR (green) and Stat2 (red). Images of cross-sections (z-stacks) of 0.17 µm thickness were acquired on a Leica SP5 confocal microscope and 3D reconstructions were generated using Imaris software (Bitplane, Zurich, Switzerland). The first of the two rotations of the 3D reconstruction shows all three channels as indicated above, the second rotation shows the blue channel (DAPI, nucleus) and the “co-localization channel” (yellow, calculated from the co-localizing signals of the red and the green channels).
