English: Tomogram of core uncoating in the cytoplasm. This movie shows a tomogram of an extended flat area of a Ptk2 cell grown on an EM grid and an intracellular core in the process of delivering the genome in to the cytoplasm. A section of the tomogram is shown in Figure 4E. Each frame of the movie corresponds to a 1.62 nm thick section through the tomogram moving along the tomographic Z-axis. The core opens up on one side through which the viral DNA is released as a whole for subsequent cytoplasmic replication. Apart from the core rupture, no further changes could be detected when compared to cores of particles attached to the cell surface. Around the core several organelles and cytoskeletal elements can be seen. |
